Genomic DNA is modified in a post-replicative manner and several modifications, the enzymes responsible for their deposition as well as proteins that read these modifications have been described. Here, we focus on the impact of DNA modifications on the DNA helix and review the writers and readers of cytosine modifications and how they interplay to shape genome composition, stability and function.
